A sensitive and reliable detection of thrombin via enzyme-precipitate-coating-linked aptamer assay.
Stable enzyme precipitate coatings (EPCs) with high enzyme loading were employed to develop a sensitive and reliable detection protocol, termed EPC-linked aptamer assay (EPC-LAA). EPC-LAA achieved the limit of detection to be 0.5 ng per mL of thrombin (13.5 pM), and maintained a reasonably stable detection signal even after storing all the reagents at 40 °C for 66 days.
متن کاملEnzyme-Linked Immunosorbent Assay of Progesterone in Serum Using Penicillinase as Label
An enzyme-linked immunosorbent assay (ELISA) for progesterone measurement in serum or plasma samples using penicillinase as label enzyme is reported. A C3 and C11 derivatives of progesterone were prepared and conjugated to bovin serum albumin (BSA). Polyclonal antibody against these two immunogens were prepared in New Zealand white rabbits. Purified Ig fractions of antibodies were immobilized o...
متن کاملSpecific detection of Shigella sonnei by enzyme-linked aptamer sedimentation assay
Development of potent new anti-Shigella agents for rapid and specific detection and treatment is of great importance. Aptamers, nucleic acid oligomers capable of specific binding to a wide range of non-nucleic acid targets, may be of value for this purpose. In the present study, we used a Systematic Evolution of Ligands by Exponential enrichment (SELEX) process to select DNA aptamers that b...
متن کاملComparison of a Time-Resolved Fluorescence Immunoassay and an Enzyme-Linked Immunosorbent Assay for the Analysis of Atrazine in Water
Immunoassays for atrazine based on a time-resolved fluorescent label and an enzyme label were optimized and utilized to measure atrazine in water. The time-resolved fluorescent immunoassay (TRFIA) was based on a polyclonal antibody and a europium label, whereas the enzyme-linked immunosorbent assay (ELISA) utilized a monoclonal antibody and horseradish peroxidase as the label. Detection limits ...
متن کاملDevelopment of a Cryptosporidium oocyst assay using an automated fiber optic-based biosensor
An intestinal protozoan parasite, Cryptosporidium parvum, is a major cause of waterborne gastrointestinal disease worldwide. Detection of Cryptosporidium oocysts in potable water is a high priority for the water treatment industry to reduce potential outbreaks among the consumer populace. Anti-Cryptosporidium oocyst polyclonal and monoclonal antibodies were tested as capture and detection reage...
